In vitro study | DAPTA is a non-toxic inhibitor of experimental antiviral invasion. DAPTA effectively inhibited the binding of cd4-dependent gp120 Bal and CM235 to CCR5 with an IC50 of 0.06 nM and 0.32 nM, respectively. In immunoprecipitation experiments, 1 nM DAPTA was able to prevent the formation of the complex of gp120/sCD4 with CCR5. By confocal microscopy, FITC-DAPTA and CCR5 were observed. DAPTA increased the release of Th2 cytokines by IL-4, IL-10 and IL-13, inducing an effective Virus suppression state in infected macrophages; both pro-inflammatory factor IL-1 and TNFα production were inhibited. Thus, DAPTA enhances the expression of Th2 cytokines, inhibiting Virus replication of macrophages. |
